The vectorology platform deals mainly with the production of retroviral vectors based on MLV, HIV-1 and SIV for multiple applications:

  • Expression or over-expression of endogenous genes or ectopic expression of genes in cell lineages

  •  Knock-down of genes by vectors encoding shRNAs

  • Regulated expression of gene or conditional knock-down of gene expression using inducible lentiviral vectors

  •  Lentiviral transgenesis for the creation of transgenic mice 

  • Genome modification using CRISPR-Cas 9

  • Production of retroviral vectors based on MLV, HIV-1 and SIV in L2 and L3 biosafety level labs

 

  • Providing assistance for:

  1. Choice of vector

  2. Cloning tips for genes of interest

 

  • Safety level classification of the vector

 

  • Virions titration: 

  1. FACS analysis

  2.  Drug resistance selection (puromycin, neomycin, …)

  3. Quantification of physical particles by Elisa on viral capsid

  4. Q-PCR

 

  • Lentiviral transgenesis for the creation of transgenic mice in collaboration with AniRA-PBES
